What Is It?

Tadalafil is a prescription oral PDE5 inhibitor approved for erectile dysfunction and for urinary symptoms of benign prostatic hyperplasia (BPH).

Claimed Active Ingredients

  • Tadalafil
    2.5 / 5 mg daily · 10 / 20 mg on-demand
    VERIFIED INGREDIENT

A claimed ingredient is what a label or seller states. It is not treated as confirmed.

04

What Can We Verify?

Established

  • Long half-life (about 17.5 hours) is well documented.
  • Efficacy and safety established in large controlled trials.
  • Dual approval for ED and BPH symptoms.

Uncertain or unverified

  • Products sold outside licensed channels cannot be assumed to contain the stated dose.
05

How It Works

Sexual stimulation releases nitric oxide (NO) in penile tissue. NO raises levels of cyclic GMP (cGMP), which relaxes the smooth muscle of the corpus cavernosum so arteries widen and blood flows in.

The enzyme PDE5 normally breaks cGMP down. PDE5 inhibitors block that enzyme, so cGMP persists and smooth-muscle relaxation is sustained. They do not create arousal—stimulation is still required.

Tadalafil's structure is cleared slowly by the liver, giving a half-life roughly four to five times longer than sildenafil's—hence a sustained window of responsiveness rather than a narrow dose window.

Benefits / Potential Advantages

  • Long window reduces timing pressure
  • Daily low-dose option
  • Absorption largely unaffected by meals
  • Also treats BPH urinary symptoms

Limitations

  • Side effects can also last longer
  • Onset can be slower than shorter-acting agents for some users

Side Effects & Precautions

Common effects

Headache, back pain, muscle aches, indigestion and flushing. Because of the long half-life, effects can persist longer.

Major interactions & warnings
  • • Nitrates (e.g. nitroglycerin, isosorbide): combined use can cause a severe, potentially life-threatening drop in blood pressure. Contraindicated.
  • • Riociguat and other guanylate cyclase stimulators: contraindicated for the same reason.
  • • Alpha-blockers (e.g. doxazosin, tamsulosin): not universally contraindicated, but combined use can lower blood pressure and requires medical guidance on dosing and timing.
  • • Significant cardiovascular disease: sexual activity itself carries cardiac load; a clinician should assess fitness before treatment.
  • • Priapism: an erection lasting more than 4 hours is a medical emergency.
  • • Sudden vision loss (rare NAION reports) or sudden hearing loss: stop use and seek urgent care.

SynMale Verdict
WELL ESTABLISHED

Strong evidence and a uniquely flexible dosing profile make tadalafil a mainstay of ED treatment.

Bottom Line

What You Should Know

Tadalafil lasts much longer than other ED tablets and can be taken daily or on demand. Food has little effect on it. The same nitrate and riociguat contraindications apply. Side effects can linger because the drug stays in the body longer.
